Antibody-drug conjugates (ADCs) are a breakthrough in drug development, fusing monoclonal antibodies with potent small molecule drugs. Widely used for treating cancer and other diseases.
At the core of an ADC are three vital elements: a monoclonal antibody, a chemical linker, and a biologically active cytotoxic payload or drug. To ensure safe handling of the potent cytotoxic payload, ProSys has ingeniously crafted custom containment isolators with a Containment Performance Target (CPT) of 1 – 10ng/m3 for ADC processing.
One key application of our containment isolators is conjugation, where the isolator facilitates the precise mixing of the potent API powder with liquid. Through a peristaltic pump, the resulting mixture is pumped to an external conjugation vessel, maintaining a seamless workflow.
Our isolators are intelligently designed, featuring a transfer chamber and a main working chamber at minimum. If space permits, a second transfer chamber enables a unidirectional process flow, maximising efficiency. We prioritise safety, which is why open transfers between the isolator and the room are strictly avoided, thanks to the incredibly low CPT requirements.
Let’s walk through the general process steps within our isolators:
- Bulk powder and liquid containers are introduced through an infeed Rapid Transfer Port (RTP) into the entry transfer chamber.
- The powder is meticulously weighed after being scooped from the container.
- Liquid is poured into a mixing vessel, and the weighed powder is added for thorough blending on a stir plate.
- Once the stirring/mixing process is complete, the solution is transferred to an external conjugation vessel via a peristaltic pump.
- After cleaning the containers and the transfer chamber, they exit through the exit transfer chamber, ensuring meticulous cleanliness.
- All chambers undergo rigorous cleaning to prevent any cross-contamination when introducing a new product.
Our high containment isolators offer an array of capabilities beyond conjugation. Solvent concentration, purification using a rotovap, tray drying, lyophilisation, and packaging of ADCs can all be safely performed within our isolators.
Built with 316L stainless steel, our isolators prioritise hygiene and easy cleaning with radiused corners. Safety glass windows with inflatable seals, HDPE inner doors with inflatable seals, and HDPE glove ports ensure seamless internal access and material passage. The isolator chambers operate under negative pressure, drawing air through a single push-push HEPA filter and exhausting it through a double push-push HEPA filter. We can even provide special environments with precise 02 and / or RH control as needed.
With Allen Bradley or Siemens PLC control and a user friendly HMI touchscreen, our isolators offer effortless operation and control. The cleaning process is optimised using spray guns, spray balls, and automated isolation valves under PLC control.
